PCR amplicons

Long-read sequencing of PCR products · about 1 minute

Clean up (optional). Purify with a column or beads, or send it unpurified and choose “DNA cleanup” when you order.
Measure the concentration with Qubit or NanoDrop. Purity A260/280 ≈ 1.8.
Check it is in range for your option. Only dilute if it is too high (EB buffer or water).
Aliquot at least 10 µl per sample into 200 µl PCR strip tubes, and write the sample number on the side of each tube.
Seal and pack. Close the caps firmly, seal them with Parafilm and wrap the tubes in bubble wrap.
Print your shipping label from the order confirmation (it carries your order number and QR code).
Bag it and ship. Put the tubes and the printed label in a zip-lock bag. Ship at room temperature: no ice packs, no dry ice.

Whole-plasmid long-read sequencing · about 1 minute

Prepare pure plasmid DNA. Miniprep with a column kit and elute in EB buffer or nuclease-free water. Purified, double-stranded, circular DNA only.
Measure the concentration with Qubit or NanoDrop. Purity A260/280 ≈ 1.8.
Check it is in range for your option. Only dilute if it is too high (EB buffer or water).
Aliquot at least 10 µl per sample into 200 µl PCR strip tubes, and write the sample number on the side of each tube.
